From Monica.DeLay at cchmc.org Wed Oct 7 10:59:41 2009 From: Monica.DeLay at cchmc.org (Monica DeLay) Date: Wed Oct 7 10:59:46 2009 Subject: [Researchflowcore] Move Announcement and Open House! Message-ID: <4ACC749D.2D61.003D.0@cchmc.org> RFCC Announcement: The FACSAria sorters and FACSCanto (3-laser) are moving to the 5th floor!!! Move Information: The move will occur on October 23rd. Access to the sorting facility will be limited on Thursday, October 22nd for packing and decontamination of instruments and will be CLOSED on Friday, October 23rd for moving. The sorters will then be tested over the weekend (24th-25th) to ensure proper function after moving and to minimize disruption of the facility. The FACSCanto should be up and running by Friday afternoon. Sorter Location: The sorters will be located in room R5001 which is down the hall from the FACSCanto room (R5529), towards the animal facility. R5001 will contain an inner room to house a sorter in a biosafety cabinet. This cabinet provides the environment to sort samples requiring BSL2+ containment (e.g. HIV, LCMV and adenovirus infected samples). The outer room will contain a sorter in a BL2 environment. FACSCanto Location: The FACSCanto 3-laser analyzer will be moved to the LSRII and FACSCalibur room (R5524) on October 23rd. This instrument has an optical layout similar to the LSRII and will be available for use 24/7 providing more overall acquisition time for analyzer use. If you have never used a Canto before you MUST complete a short training session before using this instrument. Please contact RFCC staff for more information or to schedule a training session. OPEN HOUSE: In honor of our newly consolidated core facility we will hold an open house! On Wednesday, October 28th representatives from BD Biosciences will be here for our grand opening. Mike Hulsey (Applications Specialist) will give a seminar on compensation from 10-11 a.m. and from 1-3 p.m. we will hold our open house. So come to the 5th floor in R building on Wed, Oct 28th from 1-3 to tour our new space! There will also be demonstrations of our ImageStream and Cellometer automated cell counter. The Cellometer Vision by Nexcelom visualizes cells on a slide in brightfield and 2 fluorescence channels (510-570 and 570-650nm). More information regarding this instrument will be available soon on our website under ?Core Equipment?. Monica DeLay, M.S. The Canto II had a preventative maintenance done. Please note that your voltages may change from the last time you used this instrument. Please adjust them accordingly. The Canto is having issues degassing the flow cell so the service engineer needs to return either Friday or Monday. I will let you know when as soon as I find out. He will be replacing a pump in the fluidics cart to hopefully solve this problem. In the mean time, you may still use this instrument, but be aware that if you get a bubble in the flow cell you may not be able to get it out. In addition, preventative maintenance was done on this instrument so voltage settings may be different. Thanks, Monica Monica DeLay, M.S. You will STILL call us at 636-2770 to schedule an appointment but email confirmations will be sent automatically once your information is put into the system. You will receive an email when an appointment is created, modified or canceled. In addition, there is a link at the bottom of the email for an online survey to communicate feedback on your sorts. We are VERY excited to eliminate the need for a paper calendar and feedback form and we hope that this will streamline the scheduling process. In the mean time, please be patient with us if we ask a few more questions than normal the next time you schedule a sort. We are continuing to make improvements to this system to increase efficiency. He MAY be here this afternoon, but if not, he will come tomorrow. I will let you know as soon as he confirms a time but I wanted to give the users for tomorrow a heads up if you would like to move your time to another instrument. The Canto II and 3-laser have available times still for tomorrow. Also, I deleted the reserved time for BD on the Canto for today in case anyone would like to move their time up. The SSC parameter is distorted most likely due to bubbles in the flow cell and they cannot be removed without servicing. The service engineer will be here tomorrow around 10 am to replace the pump. I will send another email when it is fixed. In the mean time, all other analyzer users, please only sign up for time that is needed and/or call the next person in line so that we can fulfill scheduled time tomorrow.
